Using VS2008, I am updating the item.ascx file created under the DV>Templates>442> folder. I have removed the table details and applied div's to gain more control. I have put in a link ref to a custom css file. I can see the style changes in VS but they are not visible on the my local test site browser? All div changes have worked?

So the DIV changes work but not the CSS? I would install FireBug and see if it shows the CSS is being pulled in.
Another option is use a module such as ‘Script Injector’ and inject the CSS stylesheets right there.
